That's something that needs to be taught in high school. Understanding probability, basic statistics, etc. is required in almost EVERY field.

PCR was never designed to be a diagnostic test.

Thats a really stupid thing to say. Anything can be a diagnostic test. PCR is actually well suited to being a diagnostic test since primers are very specific and its easily scalable.

How many threshold cycles?

Needs to be optimized depending on the application.

You like to just repeat what you've heard without actually taking the time to understand it and it makes you look like a moron. This is just the most recent. You do it like every week.

Correct assessment.

The mathematics is that the optimal is about 25.

Too low threshold cycles and the false negatives are too high.

Too many and the false positives are too high.

Either way the test has poor accuracy even at the optimal point.

Doctors are specifically trained to take this into account. Theres specific training on evaluating diagnoses in conjunction with false negatives and positives.

Really we should probably be using less sensitive tests that are faster and wont detect non contagious low viral loads. But we have what we have and its better than nothing

If he really thinks that he had better have some peer reviewed literature to back it up.

So far there has been no evidence if human interference in the viral genome, and we know what to look for so thats a pretty good indicator.
