I know Star value information is not public, but I was wondering if donations were cumulative to hit that threshold, or if you had to donate all at once to receive it.

So for example would I need to donate $100 all at once to get the "$100" star? Or could I also donate $20 in five separate transactions and also get the same "$100" star? I didn't see any specifics on the star/donation page besides them stating the screening for donor forum had to be $25+ in one donation

Yes, the values add up, you don't have to donate the entire amounts at once.
