You can only throw magical or higher items in the shrine. and only when you are missing health/mana

The well is the opposite, you can only throw non magical items (Whites) in the Well

Just adding on:

Can not shrine Maps, Glyphs & Rods.

Can not Well white rods

Fish, Comfrey, Potions, Glyphs & Totems are consumables and it gives instructions how to use them... or click and drag to the use button

What is the Well? I can't see that in the town area?
Do I need to unlock it?

As they have mentioned above, you can drop magical and above items into the shrine to help replenish life/mana - the better and higher tier the item, the more it provides - you MUST be in town and wont work if you are full
Wells can be 'found' exploring in catacombs and maps, and you can drop in white items to fight a stronger evil presence-esque mob that will drop that same item but with modifications (potential to find good loot)
