3 brothers will be in it but not major bosses im guesing.. we all ready know that:

Lilith takes a "major" role in the upcoming Diablo trilogy by Richard A. Knaak. In an IRC interview, Knaak confirmed Lilith's role in Diablo 3.

Andariel was lilith's daughter so there might be father/mothers to the 3 brothers.. im guesing there will be anyways ^^

check out the diablo look alike at the end of the D3 cinematic with 3 heads O.O
