so for diphenhydramine hcl, aka benadryl is 8.5 +/- 3.5 hours

lets just say its 8 hours. i believe the +/- 3.5 hours is for older people mostly, prob the added 3.5 hours

id assume younger is less time in system?

but my main question is

lets say u take 50 mg of it, then in 8 hours your body should be feeling the effect of 25 mg? or does the mental/physical effect of tiredness go away quicker than it takes for the half life of the plasma concentration?


like lets say if 50 mg makes me really tired... and if i took 100 mg, then in 8 hours, would i still feel really tired? cause plasma levels should be around as if i took 50 mg?

receptor desensitization and internalization predicated upon agonist/antagonist efficacy % (ligand binding affinity) & drug/receptor dissociation & idiosyncratic phosphorylation rate of that particular receptor via GRKS, and sometimes either PKA or PKC depending on the coupling. Also heavily mediated by arrestins.
that's for GPCRs, at least, of which the H1-H3 receptors are, so it fits for you.
heh, interestingly, nicotine (even though it binds to ionotropic receptors) paradoxically causes the antithesis - upregulation via diffusing directly into the soma then ER to act as a molecular, pharmacological chaperone for alph4-beta2 NAchRs. It's a funny phenomenon imo so I thought I'd share while we're on this topic.

appreciate the response, but i gotta say i dont really understand too much of that lol

the half life does vary with age because it is rapidly metabolized and is subject to many factors (level of enzymes in liver, ability of organs to clear the blood etc.)
look up "tachyphylaxis"

you would still feel tired longer into the day if you took a larger dose... you wouldn't necessarily be feeling the effect of half of it because your body would start to adjust to functioning and it would do other things like alter the levels of stress hormones (like cortisol and others perhaps) to keep you awake and functioning.

in other news - why don't you tell me why you're using this and perhaps I can suggest a therapeutic alternative which will avoid the adverse effects

difficulty falling asleep unless extremely tired

often takes 2 to 4 hours to fall asleep once lights are out

i know basic stuff like dont watch bright screen before sleep and stuff

well, if you are following those "basic things" and other non-pharmacological strategies to no avail... then perhaps speak to your doctor... there are tons of better sleep aids out there (again, I can suggest if you want and provide more info) which will no create such a strong hangover/require frequent dosing increases/reduced efficacy so quickly. This is especially true if you have insomnia.

what info are u asking of me as well?

also with diphenhydramine i do sometimes get that paradoxial effect, where i get more anxious and am fuked for the night and cant sleep even more so... sometimes not always though, seems to happen if i take it too early before i try to sleep
