Pokerus lasts for about 1hour - 2hours of time outside of a PC, it lasts forever if you keep the pokemon in a PC.

It is completely random once you catch a wild pokemon. It won't tell you it has it untill you check it up with a pokemon center and the lady says "Oh. It seems one of your pokemon have Pokerus."
