What's the point of no return? Is it after I beat all 3 graviteons and go up the tower?

when your able to enter meio's tower, it will tell you that upon entering, you can't go back.

How many health canisters are in the tower? If you remember.

So I can grab all the trophies minus the one I missed for the balrog and finishing in under 4 hours before the tower?

you sure? im pretty sure there are a couple in the tower, dont remember though

I didn't come across any up grades if can recall
