do i need a semi colon anywhere?

IMO it's an extremely awkwardly constructed sentence, but

"Barbell-benching, dumbbell-curling, lean, mean, lifting machine John is ready to hit some iron!"

would be the proper way to punctuate it.

Compound adjectives made by joining a noun and verb in gerund form should be hyphenated unless they're already well-established in the lexicon as a single word.

then there shouldn't be a comma after 'mean'

Also, the sentence fragment should be avoided considering OP is probably writing this for a formal or academic context.

At any rate, rewording it is preferable to either.
