I believe the letters represent the order of power in the organization. Juha bach said that Ishida was going to be the same as him, and that was an “A” rank. People were expecting the “B” rank to be the successor, as the rank is the next in line.
I don't think its going to be rankings because of the way the names are set up. O the Overkill, Y the Yourself, etc. etc. The A is obviously the commander because of being the first letter, but the rest might not be.
But then again it might be like the Espada where A-G or some arbitrary thing is power and everything else is name.

They're pretty obviously real quincies since its been confirmed that Juha at least was the original quincy.
