This is yet another thing that Kishimoto failed to clear up but yes, it appears as though Hashirama and Tobirama died young. We don't know how Hashirama died, I'm guessing disease, AKA a safe way to kill off extremely powerful characters (Itachi).

Im sure in his fight with madara with final susano he sustained some injuries
