I would say try Spice and Wolf. The interactions between Holo and Lawrence are hilarious, it's more of an adult setting(ie. not school), it doesn't require any knowledge of Japanese culture to understand any of the humor, Holo's dub is actually quite amazing and it was just overall a good show.

Galaxy Angel had it's moments as well. Not a great show, but decent to start with in terms of comedy.

Things that are a little more Japanese involved, maybe Love Hina though that may be borderline on the echi. All Miyazaki movies are pretty good for those new to anime.

No offense but I feel like A LOT of the reccomendations here are not relevant, well targeted, and suggestions like nichijou/gintama/nichibros are just going to give the person the feeling of of "wtf japan."

So, to make it look like I didn't come here just to critisize without anything better to say, there were a few good reccomendations people made. Here they are, I endorse because they all opens new avenues/possible interests and are beginner friendly.

Older/middle aged persons I've found usually get super turned off at violent stuff I've found, also. But can't speak for every middle aged lady o_o.

I saw it in Japanese, but I guess the dub is as good as it's gonna get.

(The movie is much better than the trailer might lead you to expect, btw)
