If you have only 1 drive then your bios is seeing the hard disk.
You OS is corrupted and you now have to use the windows 10 recovery options.

Boot from either the USB or go into the recovery console for windows 10
there you can either fresh install or install fresh but retain personal files.

/edit
After that I would test HD with HD Tune, run a deep scan followed by a benchmark and post that back here.
Before running the deep scan back up data. if the drives is dying this can kill it.

how do I go into the recovery console? I'm trying to boot from the USB but it just comes up with a blank screen with a dos cursor at the top left and does nothing...not sure why.

If we knew what motherboard you have, we could download the manual and look up your BIOS settings.

When you first enter the BIOS, it should tell you in the top-center of the screen, the model of your ASUS motherboard. We need to know that.

Also in the top-right corner you might see something like: Exit/advanced mode.

It would be nice to have the ASUS model number and have you in advanced mode.

M5A97 LE R2.0
BIOS version 1401

I can get into the advanced mode. Mouse and keyboard are working in BIOS.

should go to the advanced trouble shooting menu
from there select trouble shoot.
then advanced settings then automatic repair.

if your boot menu key is f8 disable it in the bios and try again.

Pretty sure ASUS' BIOS doesn't allow you to disable the F8 key.

On the Boot Menu, under Boot Option Priorities, it will list all the devices it finds in your comp. That is where you check to see if the hard drive is listed.

I'm on skype with him now.
the drive is detected but it won't boot the recovery console.
Either the drives bad or the OS is too corrupted.

Were going to reinstall 10 now, then test the HD.
