Runes are introduced during the Vessel of Hatred campaign quest, at the end of the Darkening Clouds storyline when you hand in the quest Fundament of Faith — there’s no minimum level or other requirements. Completing this quest will reward you a Kurast Armory Cache and a couple of random Runes.
