I have to have plugy?? in order to use ladder runewords in single player correct?

No sir, I dont use plugy and I have them active. You can try this method https://forums.d2jsp.org/topic.php?t=71588404&f=161 instructions are on reply #3

Or you can use the "ladder runeword mod"
