The number I have heard over and over is 350 + the champions/uniques/minions/cowking.

If you want to do basic math to estimate how many runs it will take to get a ber rune, it's something like 1,868 p1 full clears. (730549/391)=1868.4.

It will take slightly less on average because of the poppables and the champions/uniques/king have a better HR drop rate.

p8 would be 319615/391= 817.4 runs to get a ber rune on average.

It would be about ~7,600 p1 trav runs to get a ber rune.

The best average "value/time" per run in an area that is good for HRs is chaos sanctuary. It's less HRs/Time than cows, but it's better for uniques/rares/charms/jewels/essences. Diablo is one of 3 bosses in the game that can drop skill+45 life grand charms, which can be rerolled with pgems.

Again these are all estimates.
