I heard the audio. It had a lead in to the quote, so I really don't see how it was unscripted.

Back in chains can mean a lot of things unrelated to race.

...this is becoming the nastiest campaign in my memory . I've been going over all the ones I have witnessed in my political life and except for the rhetoric of '68 DNC no other comes close and we have 3 months to go .

I swear, every day Republicans are outraged and butthurt about some new attack by the Obama campaign. Yet the entire Romney campaign is based entirely on negative and false ads against Obama.

Stop whining. Politics ain't beanbags son.

...I have very personal reasons for objecting to the race card being played by anyone on either side especially during a Presidential election and I believe most Americans are with me on this !

I call bullshit. I can't really take you seriously unless I've heard you apply the same standards to the Republican party's use of The Southern Strategy to win elections for the past 40 years. And if you really have personal reasons to have your widdle feewings hurt from a little tough talk, you should be truly outraged and ready to give Republicans hell for voter suppression efforts in dozens of states targeting minority voters likely to vote Democratic, eh?

Later on in the day yesterday, Biden clarified that he meant to say "in shackles" rather than "in chains". It was a play on words referencing a phrase Rick Santorum often used during the Republican primary. Santorum would rail again Dodd/Frank and say we must "unshackle Wall Street". There was nothing racial about it as you already said.
