Back in my plugy days I made a plague java walkthrough here on JSP. She got lucky with a number of jav skillers and I ran LK for Bramble, Meph for Titan's etc so she was fairly kitted. To answer your questions:
1) Pretty fun in that it's something different from your usual java!
2) If we're talking /players8 then it's pretty slow going. Plague Javelin applies its damage over a much longer time than Poison Nova, for example. However with GG gear she can pump out some damage, just over quite a long time
3) Bramble with 3/20 gloves will give higher damage. If you went Enigma you'd go Trang gloves instead.
4) If you paired it with Charged Strike (see below) I'd imagine you'd aim for the same desired IAS points as regular java with Titan's, can't remember what right now
5) As Plague Javelin plus synergies is not many points, it's definitely worth having a backup skill, not least of all because running bosses with PJ is torture, and poison immunes are going to be largely untouchable. Charged Strike was a natural choice for this, you can even max a synergy or two so it's fairly poky.
The main issue I found is that she doesn't really have much to reduce enemy poison res. I guess if you had the means you could get a jmod and fill it with facets or something? Not sure. On single player, a wand with Lower Resist charges on switch is a must-have!
