If the party member's portraits are colored (not greyed out like when someone is in town) while you're outside of the room, then i think that guarantees you get the quest.
Tested as you described. Didnt work you have to be in the room.
Portrets are not grayed out. Lilly and Sin are missing and you cant go to act 6. So not hc viable
edit: and you cant go to the next act if you get the quest like the other acts. You have to talk to Lilly. access is granted after that.
Proberly because its a 2 x 5 acts storyline. It seems like 10 acts but it is theory 2 x 5.
