Lucifer wanted Abel. Cain didn't want Abel to get tempted by Lucifer. So Cain made a deal with Lucifer to protect Abel's soul. Part of the deal was that he had to kill his own brother. In turn, Lucifer now has Cain as a demon. The first blade (the bone) was the weapon Cain slew Abel with.

Cain says "When I summon you..." to Dean, suggesting a foreshadowing moment before we knew what Dean would become (it makes sense that Cain would have at least some control over the Knights). Does anyone else remember that particular part?
